研究方向

1. 多代谢组学在心脑血管创新药物的发现、评价和成药性等关键环节应用研究
以系统生物学和转化医学研究理念为指导,利用基于分析技术代谢组学对心血管疾病动物模型、临床病人及药物干预组织及生物体液代谢特点进行研究。力求在精确刻画生命体的分子特征和药物作用的物质基础前提下,探讨二者在生命体中可能的作用与影响。上述工作已发表论文19篇。
2. 蛋白质溶液结构分析及生物大分子与药物小分子相互作用研究
采用NMR技术精确测定药物小分子的结合位点,并评估药效分子群与靶蛋白的竞争结合能力,从一定程度上阐明药物小分子与靶蛋白相互作用机理。有研究包括抗阿尔茨海默病药物(BACE1抑制剂)的结构改造;新型调血脂药物候选化合物IMM-007与AMPK的相互作用;新合成的硫酸软骨素(cs) 与肿瘤坏死因子-a(TNF-a)作用关系;基于活细胞内核磁共振(on-cell NMR)技术的KRASG12C蛋白特异性结合抑制剂高通量筛选方法建立等。上述工作已发表论文3篇。
3. 复杂混合物系统微量物质结构分析
利用HPLC-NMR联用技术,鉴定了天然产物,药物杂质,生物代谢物等五十多个样品。并确定了代谢产物代谢、排泄途径,为其作用机理的阐明提供了一定依据。上述工作已发表论文13篇。
